Setting exposure with Flash

Hello, I'm new, both to photography, and this forum. This is my back story, feel free to skip to my question below:
My first camera was a Canon Ixus digital one, which I've happily been snapping away on. I work at a local college and part of my job was setting out darkroom chemicals as cover for a technician on maternity leave. I've now been shown how to develop black and white film/prints and now I'm hooked! But I've been so used to autofocus, autoexposure, that I'm only just finding my feet with manual cameras, although its certainly a fun experience!

Actual Question:
How do you expose when using a manual camera and flash? I have 2 cameras with light meters built in (Pentax K1000, and Vivitar 3000s), and I understand how to use the meter in daytime, but how does using a flash factor in? I'm going to a festival soon and I'd like to take pictures at night and I'd like them to come out

I have a lomo colorsplash flash at the moment, but I'll be borrowing a real flash

If your flash don't have a guide on the back of them, you'll have to find out the guide number and figure the exposure from there. Are you familiar with that?
